糖鎖認識接着分子、セレクチン群の機能解明と新世代抗炎症、抗癌転移薬開発への応用

研究概要

今年度は、本研究の最終年度にあたる。平成7、8年度にわたる成果として、セレクチンに対する新しい糖鎖リガンドとして、従来報告されていたSLexに加えて、硫酸化糖鎖が見出された。このうち、硫酸化されたガラクトシルセラミドすなわちスルファチドは、L-セレクチンおよびP-セレクチンに対する強力な糖鎖リガンドであることがin vitroで確認された。本年度は、セレクチンを介する動物炎症モデルの開発も行われ、コブラ毒因子投与によるラット肺炎症はL-およびP-セレクチン依存性の肺炎症であり、また、尿管結紮ラット腎炎もL-セレクチン依存性の炎症モデルであることが確立された。スルファチドは、これらのモデル動物炎症に対して強力な阻止効果を示し、新しい作用機作を持つ抗炎症薬として応用可能であることが初めて実験的に明らかにされた。また、Tリンパ球との接着によるBリンパ球の活性化にTリンパ球上のL-セレクチンとBリンパ球上のスルファチドが深く関わるという免疫学上極めて重要な知見も得ることが出来た。さらに、血流中の好中球にはL-セレクチンは存在するが、カゼイン投与などにより腹腔に誘導される好中球にはすでにL-セレクチン存在せず、刺激により極めてsheddingし易いことが示された。セレクチンは癌転移とも深く関わる接着分子であるが、本研究でL-およびP-セレクチンの糖鎖リガンドであることが明らかになったスルファチドや硫酸化糖脂質が癌転移抑制効果を持つ可能性が示された。
本研究の成果により、新しい作用機序による次世代の抗炎症薬および抗癌転移薬として硫酸化糖鎖が応用されることが期待される。
