研究概要

アラキドン酸カスケードと病態の関連性を明らかにする目的で種々の検討を行い、以下の成果を得た。
1.血小板活性化因子(PAF)に関して:PAF受容体を過剰発現したマウスを作製した。同マウスを用いた検討から、PAFは気管支喘息やエンドキシン誘発病態のみならず皮膚疾患や色素異常などにも関与する可能性を示した。
2.PG代謝産物の測定から:(1)高血圧を合併した高脂血症患者の検討から、イコサペント酸エチルは脂質代謝の改善、コレステロールやトリグリセリド(TG)の減少、さらにトロンボキサン選択的な減少効果を示し、血栓症や血管合併症の予防への有用性を示した。
(2) GC/SIMを用いた検討から、網膜血管閉塞症患者のトロンボキサン産生量が増加していること。また低用量のアスピリンの投与によりトロンボキサン/プロスタサイクリンバランスは改善され、さらに病態の改善例も認められたことから、PGバランスの変動が血管閉塞の一因になりうることが示された。
3.シクロオキシゲネース(COX)に関して:(1)マウス由来線維芽細胞における検討から、内因性アラキドン酸は、COX-2により、外因性アラキドン酸はCOX-1によりPGに転換されること。COX-1は多量の、COX-2は少量のアラキドン酸を転換すること。さらに細胞内グルタチオンペルオキシダーゼ活性の上昇に伴い、COX-2活性が選択的に低下し、低濃度のアラキドン酸は転換することができなくこと等を明らかにした。
(2)カラゲニン胸膜炎の滲出液を用いた検討から、初期の滲出液中の全COX-2の大部分は単核球と好中球に発現し、両者の寄与の程度はほぼ同等であることが示唆した。また、滲出液中の白血球のCOX-2発現レベルが低下する12時間後以降、中皮において増殖に伴うCOX-2発現を認めた。
(3)ラット腹腔マクロファージの培養系を用いた検討から、経口金製剤であるauranofinは、COX-1依存性のPGE2産生は亢進するが、COX-2の誘導を抑制することによりCOX-2依存性のPGE2産生を抑制することを明らかにした。また、COX-2タンパク質の誘導を抑制する濃度のauranofinは、iNOSタンパク質の誘導も抑制し、炎症のメディエーターとして機能するNOの産生も抑制する作用があることを明らかにした。
4.ロイコトリエン(LT)に関して:(1)抗LTB4モノクローン抗体の作製に成功し、放射免疫測定法を確立した。LTB4の生体内動態と代謝の各種病態との関連性の検討が可能になった。
(2)イオン交換カラムであるSep-pak Accell plus QMAを用いた前処理を導入した尿中LTE4のEnzyme Immunoassay (EIA)系を開発した。本法は簡便で添加回収試験も良好であった。
