蛍光色素カプセル化リポゾームによる血流、血液ガス同時連続測定法の開発

研究概要	本年度は、生体内における各種計測に適したリポソームの形状および組成を決定するとともに、その作製法を確立した。さらに、このリポソームを用いてその発する蛍光信号をin vivoにおいて測定し、血液酸素分圧を計測することが可能となった。リポソームの膜成分にcholesterolを添加し蛍光物質の保持効率を高め、さらに活性化methoxypolyethylene glycolを加えることにより、細網内皮系への取り込みを抑制することができた。作成したリポソームの微小血管における挙動を蛍光で標識した赤血球と比較したところ、リポソームの直径が8μm以下であれば、その微小循環における流速は赤血球のそれと変わらず、またリポソームの投与が原因となった血管閉塞は見られなかった。リポソームの毒性および血中半減期についても検討を加え、実験動物への応用が可能であることが明らかとなった。前年度に作成した光源は蛍光寿命を測定するには時間応答が不十分であったため、新たにアルゴンレーザーとAO (Acousto-Optic)変調器を組み合わせた光源を作成し、プラスチック光ファイバーと光電子増倍管によるフォトンカウンティングで蛍光を観測した。これを用い、まず蛍光寿命から酸素分圧を求める検量線をin vitroで作成した。次いで麻酔したラットの静脈に光ファイバープローブを装着して蛍光寿命の測定を行い、血液の酸素分圧のin vivoにおける計測が可能となった。
