肝臓内未熟樹状細胞移入による免疫学的寛容の導入と移植片拒絶抑制

研究概要	LewisラットからWKAHラットへの腎移植の系を用いて、CTLA4Igおよび樹状細胞移入の移植片生着への影響を検討した。 (1)WKAHラットの両腎摘出後、Lewisラットの腎を移植した。個体死をもって移植腎の拒絶とした。、無処理群では平均8〜10日であった。 (2)1日、100μgのCTLA4Igを7日間投与すると9例中4例に100日以上の長期生着が認められた。対照群としてヒトIgを同量、同期間投与したが、7〜8日の生存であった。 (3)Lewisの脾臓由来の樹状細胞を2×10^6個、移植前9日に静注した群では3〜6日の生存で、移植片の長期生着は認められなかった。 (4)100μgのCTLA4Igで処理した2×10^6個の樹状細胞を移植前9日に静注した群では3〜8日の生存で、移植片の長期生着は認められなかった。 (5)CTLA4Igの静注と樹状細胞の静注を別々に行う群では、19日以上の生存が認められた。現在さらに検討中である。
