アレルギー患者におけるIL-5産生の分子機構の解明-IL-5抑制剤の開発

研究概要

アトピー性疾患の好酸球性炎症成立の中心的役割を果たすCD4^+T細胞IL-5産生亢進機序を解明する目的で、ダニアレルギーの気管支喘息患者2名の末梢血より、limiting dilution法により、多数のダニアレルゲン特異的T細胞クローンを樹立した。すべてのT細胞クローンは、IL-2,IL-4を産生し、Th0型であった。抗原刺激、固相化抗CD3抗体、ホルボールエステル(PMA)+カルシウムイオノフォア(IOM)刺激に対してIL-5産生がみられるT細胞クローンにおいては、rIL-2刺激に反応して、IL-5の産生が認められた。さらに、抗IL-2中和抗体の存在によって、IL-5産生が阻止されることから、ヒトT細胞のIL-5(Th2サイトカイン)産生は、IL-2(Th1サイトカイン)依存的に行われていることが示唆された。
IL-5産生性T細胞クローン、IL-5非産生性T細胞クローンをヒトT細胞腫瘍株と細胞融合し、IL-5遺伝子発現、非発現T細胞ハイブリドーマを樹立した。ヒトgenomic IL-5遺伝子のcoding領域5′上流2kbをクローニングし、luciferase遺伝子の上流に接続した発現ベクターpIL-5.2K Lucを作製し、前述のハイブリドーマに、electroporation法にて一過性に導入した。PMA+IOM刺激によって、luciferase活性は、約1000倍の誘導がみられた。ヒトIL-5promoter/enhancer活性を観察しうるヒトT細胞として、地上に存在する唯一の細胞系が構築できたことを意味する。gel shift法を用いた核蛋白の解析では、NF-AT,AP-1,NF-κB,Oct-1の誘導には、細胞間での差は認められず、IL-5産生は、ユニークな転写因子(NF-IL5)によって制御されていることが強く示唆された。
