アレルギー患者におけるIL-5産生の分子機構の解明-IL-5抑制剤の開発

研究概要

活性化T細胞により産生されるIL-5は、好酸球活性化因子として、好酸球性炎症の成立に必須のサイトカインである。われわれはこれまでに、T細胞のIL-5産生亢進は、好酸球性炎症を主徴とする疾患の特徴であり、その抑制は、治療上の重要なターゲットと考えられることを明らかにしてきた。細胞レベル、分子レベルで、IL-5産生機構を解明し、選択的IL-5産生抑制剤が開発されれば、よりpin pointな好酸球性炎症の制御が可能になると期待される。ヒトIL-5遺伝子の転写調節機構を解明する目的で、アトピー患者末梢血からDer f2特異的T細胞クローンを樹立し、さらに、IL-5産生性および非産生性のT細胞クローンとヒトT細胞腫瘍株との細胞融合によってハイブリドーマを樹立した。IL-5産生は、ヒトIL-5遺伝子5′上流の約500bpの転写調節領域に作用して、これをpositiveに制御する転写因子によりコントロールされること、IL-5遺伝子転写能はIL-2やIL-4の転写能とは相関しないこと、また、NF-AT、AP-1、NF-kB、Oct-1の発現とも相関しないことが明らかになった。また、T細胞のIL-5産生には、IL-2とIL-4の産生に重要な転写因子NF-AT、AP-1、NF-kB、Oct-1以外のユニークな転写因子(NF-IL-5?)の関与が重要であることが明らかになった。ヒトT細胞のIL-5産生は、IL-5にユニークな転写因子によって、IL-2、IL-4遺伝子とは独立して、転写レベルで制御されることが強く示唆され、NF-IL-5インヒビターは、ステロイドに代わる夢のアレルギー治療薬として期待される。
