T細胞レセプター・クロノタイプ検出システムの臨床検査化への試み

研究概要	免疫応答の中心であるT細胞が抗原を認識する分子、すなわちT細胞レセプターに注目して、RT-PCR法とSSCP法を組み合わせることにより、リンパ球集団で集積しているT細胞クロノタイプを検出するシステムをすでに確立しており、多くの疾患において、その有用性を示してきた。今回は、このシステムをより簡単にして、数多くの検体を臨床検査のレベルで行えるようにすることを目的とした。従来、T細胞レセプターのベータ-鎖についてRT-PCRを行い、リンパ球集団におけるT細胞が発現しているほとんどのシグナルをカバーし増幅するには、20種類以上のPCRプライマーを必要としている。すなわち、一つの検体を解析するのに、20本以上のPCR反応を行い、それぞれをSSCP分析する必要がある。これが本クロノタイプ検出システムの臨床検査化を難しくしている第一の要因である。そこで今年度も前年度と同様、いくつかのコンピュータープログラムを用いて、SSCPに適した長さのDNAを増幅可能で、かつ複数のV領域のT細胞レセプターを同時にカバーできるプライマーを検策した。前年度に設定したプラーマ-は、原因は不明であるが、PCRの効率が不十分であることが判明したので、新たなものを設定した。RT-PCRを行ったところPCR産物が増幅され、そのPCR産物をSSCP法で解析したところ、従来の方法と同様な分析が可能であることが示された。そこで前年度と同様に、慢性関節リウマチ、全身性エリテマトーデスなどの自己免疫疾患患者と健常人の末梢血リンパ球における集積T細胞クロノタイプをスクリーニングしたところ、明らかなT細胞の集積を検出できることが判明し、このシステムが臨床検査のレベルで使用可能であることが示された。現在より多くの集団について検討している
