ワクチンの免疫力増強に有効な免疫アジュバントの開発

研究概要	「目的」近年ワクチンの開発は従来より用いられている死菌ワクチンや生ワクチンに加えて、遺伝子操作によるリコンビナント蛋白ワクチン、化学合成によるペプチドワクチンが行われている。しかしこれらはリコンビナントワクチン、合成ペプチドワクチンの免疫原性が弱く、免疫アジュバントの添加が必要と考えられる。私共の免疫アジュバント開発の経緯より、ワクチンの免疫原性増強のワクチンの開発を行った。「研究成果」私共はすでに20年以上にわたり、免疫アジュバントの開発を行って来たが、そのうちよりワクチンの免疫アジュバントとして有効な侯補化合物の選択を行ない、結核菌(BCG株)細胞壁骨格(BCG-CWS)、結核菌(青山B株)の糖脂質であるトレハロースジマイコレート(TDM)、ムラミルジペブチド(MDP)誘導体であるB30-MDP,MDP-Lys(L18)を選択した。治験ワクチンとして遺伝子工学的手法で作製したリコンビナントヒトB型肝炎ワクチン(rhABsAg)および山西らによって作製されたハンタウイルス死菌ワクチンを用い、マウスにおける上記抗原に血中抗体産生および細胞性免疫誘導における上記アジュバントの有効性を検討した。上記アジュバントはすべて有効性を示したが、特にBCG-CWSおよびMDP-Lys(L18)がすぐれた免疫アジュバント活性を示した。上記免疫アジュバントを抗原と共に水中油型エマルジョンとして投与した際、きわめてすぐれた効果を示した。MDP-Lys(L18)の水溶液と抗原水溶液と共に混合投与しても有効性がみとめられた。これらの免疫アジュバントのうちMDP-Lys(L18)は白血球増多剤としてすでに広く癌患者に用いられており、BCG-CWSもヒトの癌免疫療法剤としての開発がすすめられている。今後ワクチンのアジュバントとしての応答も期待される。
