研究概要	遺伝子トラップ法を用いた変異マウス作製において、トラップクローンの選択が重要なポイントなる。ES細胞の浮遊培養により得られる胚様体形成を選択法として用いることにした。まず、胚様体を用いることの是非について検討するため、内胚葉由来の肝臓で発現する遺伝子である、α-fetoprotein、transthyretin、albuminをマーカーとして用い、胚様体形成の過程での発現を解析した。その結果、α-fetoproteinとtransthyretinは早期から遺伝子が発現するが、albuminはかなり後期にならなければ発現しないことがわかった。したがって、胚様体形成を選択法として採用してもよいと推察された。トラップベクターが複数コピー組み込まれた場合は、トラップクローンからの内在性遺伝子の単離は容易ではない。このため、ES細胞に挿入された余分なベクター部分を取り除くために、マーカー遺伝子の両端にloxP配列を組み込んだベクターの作製に成功した。マーカー遺伝子としては、大腸菌のlacZ遺伝子とネオ耐性遺伝子を融合したβ-geo遺伝子を用いた。このベクターを用い、ES細胞に電気穿孔法で導入し、ネオマイシン耐性トラップクローンを100個単離した。これらを順次フィーダー細胞を除いて浮遊培養し、胚様体を形成を行なわせた。胚様体形成過程で、X-gal染色を行ない、発現パターンを解析した。その結果、基本的には6つの発現パターンに分類することができた。第1は、ずっと発現するタイプ、第2は、胚様体形成と共に、発現が消失するタイプ、第3は、胚様体形成と共に、発現が増加するタイプ、第4は、ネオ耐性ではあるもののlacZの発現は検出できないもの、第5は、中間の時期に発現が減少または消失するもの、第6は、中間の時期のみに発現が増大するものである。
