研究概要	正電荷脂質のDC-Cholを加えたHVJ-cationic liposomeが培養細胞での遺伝子発現を現行HVJ-liposomeより数十倍以上増強できた。またFITC-ODNの導入においても上記培養細胞においてはHVJ-cationic liposomeによりすべての細胞の核内に導入された。しかしマウス肝臓や骨格筋での遺伝子発現はDC-Cholesterolの入った正電荷リポソームでは殆ど認められなかった。一方DC-Cholの代わりにPSを加え負電荷になったHVJ-AVE (Artificial Viral Envelope) liposomeは培養細胞での遺伝子発現は正電荷リポソームより低いが、肝臓、骨格筋での遺伝子発現は現行のHVJ-liposomeによる遺伝子発現の5-10倍であった。蛍光オリゴヌクレオチドの組織内導入においてもHVJ-AVE liposomeは実質細胞の核内への導入に他のリポソームよりはるかにすぐれていた。このリポソームを静注すると殆ど肝臓に集積する。そこでラット腎糸球体の抗原であるThy-1に対する抗体を付けたimmunoliposomeは静注によりラット腎糸球体に集積し蛍光オリゴの腎糸球体への導入がみられた。一方腎炎の際に異常発現しているTGF-βのアンチセンスオリゴをHVJ-liposomeでラット腎糸球体に導入することにより腎炎の回復に成功した。
