超好熱菌酵素による機能性人工膜

研究概要	超好熱性細菌による機能性人工膜にとって重要なのは酵素の極度の安定性である。機能を持つ人工膜の再構成はすでに可能となり、多くのバイオセンサーが実用化された。しかしこれらの安定性は低く、長期間使用には欠陥がある。したがってa.すでに高次構造の既知な膜ATPaseと相同な超好熱V型ATPaseの機能と安定性。b.高次構造の制御を行う超好熱シャペロンの機能を解明した。a.b.共に共通のATPase相同部分があるため、クローニングには共通の方法が用いられた。 a.安定性残基の決定。超好熱菌Desulfurococcs Strain SYからV型ATPase遺伝子をクローニングし、ECGABDクラスター構造が発見された。これを発現し、電算機で相同性検索を行い、既知の好熱菌のATPaseのX線解析と対照して、その高次構造の熱安定性を残基から決定した。好熱性を高める残基置換はAsp→Glu(ヘリックス構造安定化置換)が最も多く、その逆方向(Glu→Asp)の置換は希である。次に多い置換はSer→Ala,Gly→Alaの2種の増加による内部疎水性増加による安定化であった。 b.安定性維持と再構成促進をするシャペロン類の解明。超好熱菌のシャペロニン、及びD-アスパラギン酸ラセマーゼの遺伝子構造を決定し、大量発現させてタンパク質を単離してその諸性質を解明した。超好熱シャペロニンにはαとβサブユニットがあった。なお1997年、好熱菌ATPaseのX線解析論文の共著者J.E.Walker博士(Cambridge大学)にノ-ベル賞が授与された。旧来の小さいタンパク質と異なり、高次の活性を有する膜酵素は分子量が数十万と大で、シンクロトロン放射光を用いて始めて解析できた。安定な好熱菌酵素でのみ基質の無い状態でα3β3オリゴマーが得られたからである。また、好熱菌膜酵素を用いて始めて論争の多かった分子内回転に決定的な直視観測が可能となって、回転説を提案したP.D.Boyer博士(California大学)にも今回のノ-ベル賞が授与された。本計画終了に当たりBoyer博士が本学で講演する。
