生物学的作用解析用高磁場発生装置の開発と個体〜遺伝子レベルでの磁場効果解明

研究概要	1.冷却灌流装置とビデオ監視記録装置付き交流高磁場発生装置の作製に成功した。1200V、60Hzで最大ピーク値5テスラの交流高磁場が得られ、90l/minの冷却水灌流により±0.5℃以内の精度で磁場空間を生理的温度に保つことができた。また、次年度計画の遺伝子複製・転写実験ではラジオアイソトープ標識ヌクレオチドを用いることが必須となるため、放射線管理区域内で使用可能な冷却装置付き小型高磁場発生装置も試作した。この装置は、励磁電流200V、192Aのとき実験空間70×100×6mm^3において周波数60Hz、波高値1テスラ、均一度2%以内の磁界を発生しうる。 2.種々の発生段階に同調培養した線虫にピーク値1.7テスラの高磁場を印加した。その結果、孵化後の体長増加と産卵開始時期が非磁場印加群に比し遅延する傾向が認められたが、いずれの形質も磁場解除から一定時間後には正常に復した。器官形成には異常は認められなかった。また、hermaphroditeの産卵数は磁気線量に依存して軽度減少することが見い出された。 3.遺伝子複製・転写反応のための鋳型DNA・プライマーDNAを化学合成しHPLCで精製した。 4.なお、本研究遂行の過程で、ミドリムシが強い勾配磁場中で高磁場方向に走化性を示すことも発見された。
