生物学的作用解析用高磁場発生装置の開発と個体〜遺伝子レベルでの磁場効果解明

研究概要	本研究は、交流高磁場発生装置の試作に成功した金沢大学工学部のグループと線虫という磁場効果解析に最適と考えられる生物研究ならびに遺伝子研究に実績のある同大学医学部のグループが協力して行う学際研究で、現代生活で曝露の機会が増している低周波交流高磁場の生体への影響を解明する目的で遂行された。本年度研究は当初計画通りに順調に進行し、以下の結果が得られた。 1.線虫にピーク値1.7テスラの交流高磁場を印加すると、(a)咽頭筋のポンプ運動回数が減り摂食が低下した。(b)蛇行運動と(c)飼料への走化性も減弱し、通常では認められない逆行運動や運動鈍麻が観察された。しかし、(a)-(c)のいずれの変化も一過性で可逆的であった。初年度から数えると、述べ1万体を超す線虫個体が高磁場に晒された。しかし、致死あるいは早老を示した個体は1例もなく、また、表現型で見るかぎり突然変異、復帰変異とも認められなかった。したがって、線虫は1.7テスラまでの変動高磁場には耐性をもつと考えられた。 2.試験管内DNA複製反応におよぼす磁場の影響を検討した結果、(a)DNAポリメラーゼによるDNA合成速度および(b)DNA複製エラー率は1テスラまでの交流高磁場印加・非印加で差がなかった。後者に関しては、校正エキソヌクレアーゼ活性を欠くポリメラーゼを用いてもエラー率に変化はなかった。(c)1テスラ変動高磁場は、がんや神経疾患の発症と関連づけられているDNAの滑り複製にもほとんど影響をおよぼさなかった。したがって、DNAの酵素的合成は、変動高磁場中でも反応の遅滞やエラーを招くことなく、安定に進行しうると考えられた。
