生物学的作用解析用高磁場発生装置の開発と固体-遺伝子レベルでの磁場効果解明

研究概要	本研究の目的は、交流高磁場発生装置の試作に成功した金沢大学工学部のグループと線虫という磁場効果解析に最適と考えられる生物研究ならびに遺伝子研究に実績のある同大学医学部のグループが協力して行う学際研究で、現代生活で曝露の機会が増している低周波交流高磁場の生体影響を解明することである。本年度研究は当初計画通り順調に進展し、以下の成績を得た。 1.線虫個体の寿命の長さを高磁場暴露(最大ピーク値1.7テスラ)群と非暴露群で比較した結果、両群間で有意な差は認められなかった。また、高磁場印加は変異形質および復帰変異の出現頻度にも影響を与えなかった。 2.(1)レポーター遺伝子(lacZ)にミスマッチ変異を有するファージDNAをrepair-proficientなヒト細胞からの抽出物と高磁場の存在・非存在下でインキュベート後、repair-deficientな大腸菌変異株(mutS)に移入してXgalを含む培地にプレーティングし、形成される青色プラークと無色プラークの比率から修復効率を算定した結果、高磁場印加の有無で差がなかった。(2)ポリdAを鋳型にRNAポリメラーゼによる試験管内転写を高磁場の存在・非存在下で行った結果、RNA合成の速度・忠実度に差は認められなかった。(3)熱ショックプロモーター下流にlacZ遺伝子をもつトランスジェニック線虫に高磁場を印加すると、レポーター遺伝子の発現が誘導される傾向が観察された。したがって、個体の老化・変異およびミスマッチ修復・RNA転写反応は変動高磁場の影響をうけにくいと考えられたが、磁場環境は生体ストレスとなりうる可能性が示唆された。
