ヒトのリスク推定に資する太陽光紫外線曝露線量計の開発

研究概要	我々はニトロセルロース膜上に固相吸着させたDNA中に太陽光紫外線によって誘発される損傷を定量的に検出することによって、太陽光中の有害紫外線線量を測定する系を開発することを試みた。測定には損傷特異的抗体TDM-2を用い、2次抗体に標識された酵素と基質の発色を定量化することによって紫外線被曝量を推定した。また、ニトロセルロース膜をフィルムでシリカゲルとともに密閉し、温度の変化に対応できる形態とした。オリエル社の太陽光シミュレーターを用いた照射実験では、発色ある線量域を超えてしまうと頭打ちになることが明らかとなった。紫外線をカットできるブルーポリエチレンフィルターで密封した結果、より高線量域まで測定が可能となった。また、このフィルターの透過性は温度に依存しないことを確認した。さらにDNA中に生成する損傷量は照射時の温度に依存しないことも確認した。紫外線照射後生成したDNA損傷は照射後さかのぼって22日間まで安定に保持され、保存によって損傷量が減少しないことが明らかとなった。また、DNAをニトロセルロース膜に吸着後、21日間まで保存しておいても照射によって生成する損傷量が低下しないことも確認した。以上の基礎実験から、屋外での使用に耐えうる程度に温度・湿度に対して安定であり、しかも作製してから少なくとも3週間程度は安定であり、太陽光曝露後も3週間はさかのぼって測定ができる測定系であることが判明した。しかも、試作した太陽光紫外線フィルムバッジは大きさが7×3cm程度の薄い封筒状の形態をしており、携帯に簡便なように作られている。以上より十分に実用に耐えうる太陽光紫外線フィルムバッジの確立に成功したと考えられる。
