末端酸化酵素選択的抗菌剤の開発と基質酸化機構の生化学的解析

研究概要

1)新規の末端酸化酵素選択的抗菌剤を開発・スクリーニングするために、ユビキノン、アンチマイシンおよびカプサイシン誘導体の系統的合成を行った。2)阻害剤作用部位の分子構造を解明するために、bo型ユビキノール酸化酵素のみを発現する大腸菌株を用いてベンゾキノンおよび置換フェノールの耐性変異株を取得し、阻害スペクトルを明らかにするとともに変異部位がサブユニット2の膜貫通領域近傍に存在することを示す予備的な結果を得た。3)大腸菌bo型ユビキノール酸化酵素の高親和性基質結合部位(Q_H)が2電子還元を受け、また、セミキノンラジカルを安定化できることを明らかにし、基質酸化部位から低スピンヘムへbヘの電子移動を媒介できることを示した。4)活性評価システムを用いて、野生型酵素、Q_H部位欠失酵素、および金属中心欠失酵素における反応機構を解析し、Q_H部位が分子内電子移動に必須であることを示した。5)ユビキノンおよびアンチマイシン誘導体を用いて、大腸菌チトクロムbo型ユビキノール酸化酵素およびbd型ユビキノール酸化酵素の基質結合部位の構造活性相関研究を行い、また、抗菌活性を持つことを見い出した。6)ユビキノン誘導体およびカプサイシン誘導体を用いて、大腸菌グルコース脱水素酵素とウシ心筋NADH脱水素酵素のキノン還元部位の構造特性の差異を明らかにした。7)プロトンポンプ能を持つウシ心筋、ジャガイモおよび大腸菌のNADH脱水素酵素とポンプ能の無いNADH脱水素酵素を用いて、カプサイシン誘導体の阻害活性を検討した。
