生体分子エンジン素過程の高速顕微画像解析システムの開発

研究概要	(1)温度パルス顕微鏡法()に高速顕微画解析システムを組み合わせ、温度の上昇・下降速度を画像解析したところ、緩和時間は、上昇・下降ともに約6msであることが分かった。一方ガラス表面に直径10μmの円状に金属(アルミ)を蒸着し、その部分に赤外光を照射することによって蒸着金属面を熱源とする温度パルス法では、温度分布の再現性は第一の方法に比べて高いが、緩和時間は数十msと遅かった。そこで第一の方法をアクトミオシンモーターの滑り運動系に応用し、温度上昇と滑り運動との関係を解析したところ、約6ms遅れて滑り運動が始まることが分かった。この時間遅れがモーターのどのような状態に対応するかの解明は、今後解決すべき問題として残された。主要な結果は論文としてまとめ、現在投稿中である。 ():光学顕微鏡の試料面に撒かれた金属粉末の塊(5μm程度の大きさ)に赤外レーザー光(波長約1μm)を照射し、この金属塊を熱源として半径約数十μmにわたって同心円状の温度分布を得る方法。 (2)さらに温度パルス顕微鏡法を、アクチンミオシン系だけでなく微小管-キネシン系に応用した。その結果、18℃から約40℃に上昇すると、滑り速度は約0.4μm/sから約4.0μm/sへと上昇した。4.0μm/sという滑り運動は、これまで知られていたキネシン分子モーターの滑り速度の数倍にも達する。キネシン分子モーターの機能は、ミオシン分子モーターに比べて温度に対して弱いことも分かった。また、キネシン分子モーターは一分子でも滑り運動をすると言われているが、我々は一分子のレベルでも、キネシンの滑り運動機能が熱的に励起されることを確認した。従って、滑り運動機能の熱励起の研究は、温度パルス顕微鏡法と、高速カメラの組み合わせによって初めて可能になることが確認された。
