研究概要	SM/JとA/J系を祖先系統とする26系統からなるSMXAリコンビナント近交系(RI)を確立し、受精卵凍結保存を行う一方で、マイクロサテライト以外にRLGS法を用いたDNAマーカーにより系統間分布表(SDP)を拡充し、RI系統間に存在する体重、臓器重重量、血液性状、血液生化学値、肺腫瘍等における特性の違いを明らかにしていくための基礎調査を行った。 1.SM/J,A/J及びRIの26系統の兄妹交配による系統維持及び実験供試用マウスの拡大生産を行いながら以下の形質について基礎調査を実施した。 (1)体重及び臓器重量:70日齢にて体重測定後、麻酔死の後解剖し、脳、肺、心、肝、腎、副腎、脾、生殖器の重量を測定した。 RI各系統雄10匹、雌10匹を目標に件体数を増やしているところである。 (2)血液性状:各RI系統について70日齢において、赤血球数、白血球数、ヘマトクリット値、ヘモグロビン濃度の測定を行い遺伝的解析用コンピューターソフト、マップメーカー/QTLを用いてQTL解析を実施し、各形質を支配する量的遺伝子が多数あり、それらを染色体上にマッピングした。 (3)血液生化学値:各RI系統について70日齢において、血清脂質(中性脂肪、総コレステロール、リン脂質遊離脂肪酸)を測定した。 2.SM/JとA/Jの間でRLGS法によるDNAの多型を検索し、合計400個のDNAマーカーからなるSDPを確立した。 3.各RI系統の受精卵凍結保存の方法を検討した結果緩慢凍結法による保存を行うことにした。 4.肺腫瘍抵抗性のSM/Jと高感受性のA/J系との間のRI系におけるウレタン誘発肺腫瘍発生率を調べた結果SMXA24系は最も強い抵抗性を示したので、A/JとのF1、戻し交配世代についてウレタンによる肺腫瘍を誘発し、抵抗性を示した個体についてマイクロサテライト法による遺伝解析を行った。
