パッチクランプ法に適した哺乳動物網膜スライス標本の開発

研究概要	脊椎動物網膜の視覚情報処理機構を理解するためには、網膜を構成する各細胞の膜のイオンチャネルの性質、シナプス入出力、伝達物質受容体の性質の解明が必要である。イオンチャネルや受容体に関しては単離した網膜細胞にパッチクランプ法を適用することによって多くの成果が挙げられている。しかし、単離細胞はシナプス結合の解析には適さず、対象とするシナプスとその周辺に存在する細胞群を直接観察しながら、応答を記録できるスライス標本が最適である。本研究の目的は、哺乳動物網膜にパッチクランプ法を適用することができるスライス標本を作製する技術を開発することである。網膜組織は非常に薄い(約200μm)ため、市販のビブラトーム等の使用は困難である。従って、本年度は任意の厚さのスィス標本を作製できる網膜専用スラィス作成器を本学・研究機器開発室と協力して開発した。また、生理学実験のためには100μm-200μmの厚さにスライスした標本を記録槽に移さねばならない。そこで、スライス標本作成部と記録槽が接近しており、薬物潅流実験も可能なように、スライス作成部と記録槽が一体化し、且つ顕微鏡のステージ上にも装着可能なチャンバーを作成し、これを用いて予備実験を行った。キンギョ網膜を剥離し、ミリポア濾紙に張り付け、上記のスライス標本作成器でスライス標本を作製した。スライス標本は顕微鏡下で観察すると3層の細胞層が確認できた。割断面に露出した網膜細胞からはパッチ電極で記録を得ることに成功した。
