研究概要

○リパーゼをもちいたエステル交換反応における立体選択性の研究
加水分解酵素は基質に対する許容性が広く、有機溶媒中でも酵素活性を発揮するなど、有機合成に利用するのに適した酵素である。しかも、安価で入手できるものの種類が多いことも見逃せない利点である。加水分解酵素のうち、特に、入手容易なリパーゼQLを触媒として、有機溶媒中でのエステル交換反応で有機合成のキラルシントンとして有用なアルコール類の光学分割を行い、反応の立体化学を研究した。これをもとにリパーゼQLの立体選択性を活性部位モデルとして定式化した。この経験則はリパーゼQLでの光学分割が可能なアルコールを判断し、生成物の立体配置を推定するのに有効であり、このような経験則の構築はその有機合成のキラル触媒としての利用価値を高めるものである。
○光学活性クラウンエーテルの合成とキラルアミンとの錯形成におけるエナンチオマー選択性
光学活性クラウンエーテルとキラルアミンとの錯形成におけるエナンチオマー選択性の機構解明は酵素-基質複合体でのキラリテイー認識機構を知るうえでの重要な知見を与えてくれる。本研究では中性アミンとの錯形成を調べるのに都合の良いフェノール性クラウンエーテル数種を合成しキラルアミンとの錯形成における不斉識別を調べた。
化学的、生化学的プロセスでの不斉合成やエナンチオマー選択的反応で複合体でのキラル認識の過程が関与する場合、従来は、キラル識別の効率は温度が低いほど高くなると言うのが常識とされてきた。本研究ではクラウンエーテルとアミンとの錯体形成でのエナンチオマー選択性は温度によって変化することを見いだした。特に、エナンチオマー選択性がある温度で逆転し、それ以上の温度では温度が高くなるにつれてキラル識別の効率が良くなることは画期的な知見である。この現象は人工化合物でのホストーゲスト錯形成に限ったことではなく、酵素-基質複合体が関与している酵素反応でもありうることであり、酵素反応を有機合成に応用する際には不斉認識の温度による変化を十分に考慮するべきであることを示唆している。
