遺伝子標識法を用いた植物土壌病原菌のモニタリングシステムの開発

研究概要	本年度の研究においては、導入遺伝子の細胞・組織化学的検出法の確立とモニタリングへの適用を検討した。まず、前者の研究課題においては、導入する標識遺伝子として、抗生物質であるハイグロマイシンの無毒化遺伝子、β-グルクロニダーゼ(GUS)生産遺伝子およびルシフェラーゼ生産遺伝子を用い、これらの遺伝子をベクター内に組み込んだプラスミドを作製し、病原菌類細胞で機能的に発現しうることを明らかにした。初年度の研究において、遺伝子導入法が確立されたので、その遺伝子の発現を多面的に検出するシステムを検討した。検出の方法としては、遺伝子から転写されたmRNAを検出する方法をとり上げ、標識したプローブによる細胞内ハイブリダイゼーションズを行った。この場合、ハイブリダイゼーションに用いるプローブを異なる蛍光色素(ロ-ダミン、フルオレッセイン等)で標識したので、複数のmRNAを同時検出することが可能であった。以上の結果は、本年(1996年)の日本植物病理学会報第62巻576頁から579頁に発表した。また、本法の詳細については、全国農村教育協会から刊行される著書『フザリウム病』において、『遺伝子標識法』として執筆した。一方、モニタリングシステムの構築に関する研究課題においては、病原菌がある種のインドール類縁化合物に対して異なる感受性を示すことを明らかにし、同じく日本植物病理学会報の第62巻414頁から417頁に発表した。
