研究概要	植物細胞では、主要なカルシウムプールは液胞である。液胞内Ca^<2+>濃度は数mM、細胞質Ca^<2+>濃度は0.1-0.2μMと報告されている。液胞へのCa^<2+>能動輸送には、Ca^<2+>-ATPaseとCa^<2+>/H^+の2つの装置が関与している。前者はATPのエネルギーを利用し、後者は液胞膜H^+-ATPaseとH^+-pyrophosphataseが形成する△pHを利用する。ヤエナリ胚軸から液胞膜を調整し、^<45>Caを用いたメンブレンろ過法で輸送体の酵素学的性質を明らかにした。 Ca^<2+>-ATPaseは、ATPのみを基質とし、1mMのATPで最大活性が得られた。膜小胞への取込み活性はH^+イオノフォアであるCCCPで阻害されずA23187で強く阻害される。また、バナジン酸により部分的に阻害された。酵素の最大反応速度は6.9nmol/min/mg protein、Ca^<2+>に対するKmは2.6μM、ATPに対するKmは42.9μMであった。一方、Ca^<2+>/H^+対向輸送体の活性は、CCCP、A23187で完全に阻害された。基質濃度-活性の曲線はS字を示し、Ca^<2+>低濃度では活性は低く、10μM以上で高い活性を示した。最大反応速度は21.2nmol/min/mg protein、Ca^<2+>に対するKmは24.5μMであった。本研究は、ヤエナリ胚軸の液胞膜にCa^<2+>-ATPaseとCa^<2+>/H^+対向輸送体の2つのカルシウム能動輸送系が存在することを明らかにした。さらに、Ca^<2+>低濃度ではCa^<2+>-ATPaseの機能が高く、高濃度Ca^<2+>の条件ではCa^<2+>/H^+対向輸送体が高い活性を示すことを明らかにした。実際の細胞において、細胞質Ca^<2+>濃度を低く保つ役割において、対向輸送体は大量輸送体として機能し、Ca^<2+>-ATPaseは微調整をする役割を担っていると考えられる。現在、Ca^<2+>/H^+対向輸送体のcDNAクローニングを進めており、間もなく一次構造が明らかになる。
