有毒渦鞭毛藻の分子プローブによる超高感度モニタリング法の開発

研究概要

麻痺性貝毒の原因藻である渦鞭毛藻Alexandrium属は形態的に酷似し、原因藻の分類同定に混乱が生じている。そこでこれまでの研究で作製したAlexandrium spp.の外殻鎧板に特異的に反応するモノクローナル抗体により、A.tamarenseとA.catenellaの識別や現場試料への応用を試みた。また、rRNA遺伝子のITS(インターナルスペーサー)領域にDNAプローブを作製し本藻モニタリングの基礎を確立した。本年度得られた結果は以下の通りである。
(1)モノクローナル抗体M8751-1はA.catenella細胞よりもA.tamarense細胞に5〜10倍強く反応し、蛍光顕微鏡およびフローサイトメーターにより効率よく識別、定量が可能であった。
(2)抗体M8751-1は現場海域で発生しているA.tamarenseの細胞に特異的に反応し、蛍光顕微鏡およびフローサイトメーターにより検出可能であることが明らかになった。しかし、A.catenellaの現場サンプルには反応性が低く、反応性を増大させる必要が示唆された。
(3)A.tamarenseとA.catenellaのITS領域に各々特異的な蛍光DNAプローブを作製し反応特異性を調べたところ、catプローブはA.catenellaにtamプローブはA.tamarenseのゲノムDNAにのみ反応し、有毒な両種の識別が可能となった。
