家畜の糞線虫の消化管からの排除における粘膜型肥満細胞の役割

研究概要	イヌの糞線虫(Strongyloides stercoralis)、ウシの糞線虫(S.papillosus)、ブタの糞線虫(S.ransomi)の成虫をW/Wマウスと対照の+/+マウスの十二指腸に外科的に移入し虫体の排除に掛かる期間を比較したところ、S.stercoralisの排除には肥満細胞欠損のW/Wの方が+/+マウスより明らかに長期間を要した。一方、S.papillosusやS.ransomiの場合には両マウス間で著名な差がみられず、きわめて短期間のうちに虫体が排除された。このうちS.papillosusについてはマウス以外の齧歯類でも調べたところ、スナネズミやハムスターにおいてよく定着することがわかった。しかし、定着後に宿主消化管の麻痺性イレウスを引き起こし、宿主が死亡することから、現在その原因を究明中である。また同時進行していたスナネズミ腸管粘膜肥満細胞のトリプターゼ(gMCT)のcDNAクローニングに成功した。cDNAは810bpのオープンリーディングフレームを有し全長1219bpであった。gMCTのmRNAの発現をスナネズミの組織別に見ると、腸管粘膜に最も多く、皮膚や舌の筋肉中には非常に少なかった。上述のようにS.papillosusやS.ransomiの排除には肥満細胞以外の別の機構が介在している可能性が示唆された。我々の今までの研究結果からネズミ糞線虫もハムスターから排除されるときには杯細胞のムチンにより排除され得ることがわかっている。このように、同種の寄生虫でも宿主が異なるとエフェクターが本来と異なる利用のされ方をする可能性があることから、今回のように通常の宿主-寄生虫の組み合わせ以外の寄生虫を移入した場合、宿主が別の機構を動員して寄生虫を排除している可能性が考えられる。
