ステロイドホルモンの生合成と作用機構の細胞化学的解析

研究概要

1.副腎皮質束-網状帯細胞には1型5αリダクターゼが存存し、これは去勢によって発現が増え、テストステロンやエストラジオール投与によって抑制されことがわかった。副腎皮質のステロイド合成および代謝は性ステロイドの調節を受けていると考えられる。
2.副腎髄質では、神経線維を取り巻くシュワン細胞の細胞質や、髄質細胞を取り囲む支持細胞の細胞質に1型5αリダクターゼが存在する。これらの細胞における酵素の発現は去勢などの影響を受けず、性ホルモンの調節を受けていないと思われる。
3.末梢神経系では、シュワン細胞の細胞質、神経節の外套細胞の細胞質などに1型の5αリダクターゼが存在する。
4.中枢神経系では、主としてオリゴデンドログリアの細胞質に1型の5αリダクターゼが存在する。特に、髄鞘形成期に一致して発現するので、この酵素との関係が示唆された。
5.ヒトの精嚢、精管などについて、2型5αリダクターゼの局在を免疫組織化学的に調べたところ、上皮の基底細胞の他、間質のすべての平滑筋細胞の細胞質に陽性反応があり、前立腺と同様の存在様式を示すことがわかった。
