移植片拒絶反応のメカニズム:自己-非自己の認識と非自己傷害機構の分子生物学的研究

研究概要

C57BL/6マウスの腹腔内にBALB/cマウスで継代した線維肉腫細胞(Meth A)を移植すると、移植局所に2種類のcytotoxicな細胞が浸潤した。移植したMeth A細胞を傷害する主たる細胞は、マクロファージ系,非T,非NK細胞であり、移植後4,5日で移植局所に出現した。本細胞の浸潤から遅れること1〜2日後には、移植したMeth A細胞と同じ組織適合性抗原を持つBALB/cリンパ芽球にcytotoxicなT細胞が浸潤した。これら2種類のcytotoxicな細胞の浸潤と共に、移植後増殖し続けたMeth A細胞は、7日後をピークにその後減少し、約2週間後には移植局所から完全に消失した。しかし、Meth A細胞移植前に細胞毒素を含むリポゾームで特異的に貪食細胞をあらかじめ除くと、移植Meth A細胞は、増殖し続け、動物は死亡した。これらの結果は、マクロファージ系細胞が、非自己移植細胞を非自己と認識し、傷害していることを示唆している。これらの結果を基に、移植の典型例である皮フ移植の系で、移植片の遊離細胞を標的細胞に、移植片拒絶部に浸潤するレシピエントの白血球細胞のエフェクター細胞として用いたところ、本エフェクター細胞の主たるものが上述のマクロファージ系細胞であり、本細胞が、皮フ移植片を特異的に傷害していることが明らかとなった。
