ホルモン依存性プログラム細胞死の分子機構解析とその応用

研究概要	1 研究目的本研究は、プログラム細胞死を脂溶性ホルモンにより自由にコントロールしうるシステムを作りだし、さらに本システムをもちいてプログラム細胞死の分子機構を明らかにすることを目的としている。 2 研究成果 fas抗原受容体のプログラム細胞死に必須な蛋白領域とエストロジェンホルモン受容体もしくはレチノイン酸受容体のリガンド結合領域を結合した融合蛋白質をコードする融合遺伝子を作出し、fas抗原に感受性があることが知られているL929細胞及びHeLa細胞に恒常的に発現するように導入した。得られた細胞株に、導入した融合遺伝子の受容体部分に対応するリガンド(エストラヂオール、レチノイン酸)を加えたところ、どちらの細胞においても0.1nMのエストラヂオール、10nMのレチノイン酸で処理後約12時間ですべての細胞が死滅することが観察され、脂溶性ホルモンによりプログラム細胞死を自由にコントロールしうるシステムの確立に成功した。一方、プログラム細胞死にかかわる細胞内シグナルトランスダクションに関わる生化学的な解析は、現在までほとんど進展していない。我々は、プログラム細胞死のシグナル伝達の分子機構を明らかにする目的でfas抗原受容体のデスドメインと相互作用をする分子をイ-ストのtwo hhybrid法を用いて検索した。その結果、マウスfas抗原受容体のデスドメインと相互作用を示す分子をコードするcDNAを複数分離した。いずれの分子も、これまでに報告のない未知の分子で、細胞死のシグナルを伝えない変異体(C末端の欠失・lprCG変異体)とは反応を示さなかった。今後、これらの分子が細胞死に与える影響について解析を行う。
