ウイルスと細胞との膜融合を誘発する細胞性プロテアーゼの発見とその制御因子の解析

研究概要	外膜糖蛋白を持つウイルスが細胞に感染する過程は、ウイルスが細胞膜上のAdhesion moleculeに結合する過程とウイルス膜と細胞膜が融合しウイルス遺伝子が細胞内に導入される2段階に分けられる。我々はこれまでにインフルエンザウイルスやセンダイウイルスの外膜糖蛋白を限定分解し膜融合領域をアミノ末端に発現させ、ウイルスの膜融合能を発現させる臓器特異的プロテアーゼ(Tryptase Clara)を気道の粘膜上皮に見出してきた。さらにHIVについては、ウイルスがCD4レセプターに結合した後の膜融合プロセスに関与すると推定される細胞膜結合型プロテアーゼ(Tryptase TL_2)をヒトT細胞に見出してきた。本年度研究において、Tryptase Claraの活性を抑制しインフルエンザウイルス感染を抑制する生体内物質の単離に成功した。一次構造決定の結果、この物質は従来エラスターゼの阻害剤として報告のあったAnti-leukoprotease inhibitor(ALP)と同一物であると判明した。ALPは細胞培養系および感染動物実験モデルにおいて、著明な抗インフルエンザウイルス感染作用と抗センダイウイルス感染作用を示した。さらにTryptase Claraに対するALPの阻害作用は、C末端側の半分の領域に局在することが明確となった。現在C末端側のペプチドの種々の偏位体を作成して活性基の同定と、活性の強いALPの作成を試みている。一方Tryptase TL_2(TL_2)の機能解析の結果、TL_2はHIVの外膜糖蛋白質GP120と結合するが、結合後のこの蛋白をV3領域でのみ特異的に解裂することを見出した。TL_2のプロテアーゼ作用は、GMPやAMPで活性化を、GDPやADPで阻害を受けた。またTL_2はプロテアーゼ作用以外にATPase作用を持ち、蛋白質との相互作用でATPase作用が増強されることが判明した。これらのことからTL_2はプロテアーゼとして作用すると同時に分子シャペロンとして膜融合や物質の輸送にかかわっていることが推定された。
