Fas/Fasリガンドシステムの生理的、病理的意義の解析

研究概要	我々はこれまで、FasとFasリガンド(FasL)の遺伝子を単離し、これらが細胞にアポトーシスの信号を伝達する受容体とそのリガンドであること、免疫系のホメオスタシスに重要な働きをしていること等を示してきた。これらの成果をふまえ、本研究ではFas/FasL系の生理的、病理的意義を解明すること目的とし、本年度は以下の成果を得た。1.様々な分化、活性化段階のマウスT細胞の組換えマウスFasLに対する感受性を検討した。胸腺T細胞は最も未熟なCD4^-CD8^-期には耐性であり、CD4^+CD8^+期に感受性が最大になり、最も成熟したCD4^+CD8^-及びCD4^-CD8^+細胞は中等度の感受性を示した。抗Fas抗体を用いた実験から、末梢T細胞はFas誘導アポトーシスに対して耐性と考えられてきたが、実際にはFasLに対し感受性を示した。また、末梢のナイーブT細胞は、抗TCR抗体による刺激で、FasLに対する抵抗性を獲得することを発見した。これらの結果は、FasLによる免疫系の調節機構を理解するうえで、極めて重要な発見である。2.大量調製したヒト可溶性FasLをハムスターに免疫し、抗ヒトFasLモノクローナル抗体を樹立した。これらの抗体の幾つかはヒトFasLに対する中和活性を示し、またフローサイトメトリーに有用であった。3.二種類の抗ヒトFasLモノクローナル抗体をもちいて、ヒトFasLに対する高感度なサンドイッチELISAの系を確立した。この系を利用して、様々な患者血清中のFasLを定量した結果、LGL白血病やNKリンパ腫の患者血清中に多量のFasLが検出された。これらの患者は、しばしば低好中球血症や肝障害を併発するが、これらの合併症にFasLが関与している可能性が示唆された。4.マウスに可溶性ヒトFasLを投与したところ、大量(500μg)の経静脈投与で劇的な肝障害を起こして全例死亡したが、毒性は予想より低かった。しかし、P.Acnes死菌でマウスを前処理することにより、感受性は約20倍に上昇した。
