研究概要	細胞貫通型の接着分子CD44は、コラーゲン、フィブロネクチン、ヒアルロン酸等の細胞外マトリックスや細胞間接着に関わっていると考えられている。19のエクソンからなり、mRNAでは通常エクソン6〜15(バリアントエクソン)がスプライシングを受ける。しかし進行癌では種々の部位でスプライスされた多種類の異常mRNA(アイソフォーム)が存在し、転移性との関連が注目されている。本研究ではCD44の異常発現と大腸多段階発癌との関連を明らかにすることを目的とし、腺腫、早期癌、進行癌および癌転移巣を含む大腸腫瘍60検体に発現されるCD44バリアントエクソンを解析した。poly(A)RNAを鋳型としてCD44エクソン3と16-17のプライマーを用いRT-PC法によりcDNAを増幅させた後、電気泳動により分離し、各cDNAバンドに含まれる転写領域をエクソン5〜15の11種のプライマーを用いてPCR法により調べた。正常粘膜には標準型のみが発現されていたが、腺腫ではエクソン12〜15を含むアイソフォームと10〜15を含むアイソフォームが検出された。進行癌ではより大きいサイズのmDNAが検出され、卵巣転移や脳転移を示す大腸癌にエクソン7〜15を含むアイソフォームが存在していた。また、腫瘍の悪性度が増すほどアイソフォームの割合が高くなり、正常mRNAが減少する傾向が見られた。しかし、腹膜播腫を示す癌では標準型に近いサイズのmRNAが検出され、このアイソフォームに含まれるエクソンの解析を行っている。以上の結果は大腸腫瘍におけるCD44遺伝子の発現が極めて多様であることを示している。
