HTLV-I Taxと結合するRNA結合分子と転写因子の作用

研究概要

HTLV-I Taxは細胞側の種々の遺伝子の発現に影響を与え、T細胞を腫瘍化すると考えられている。その作用は、Taxが細胞側の複数の転写因子に結合し、転写を活性化するために起こる。我々は、Taxが転写因子以外に機能の不明なRNA結合分子に直接結合することを見い出し、その遺伝子のクローニングを進めている。また、ATL細胞に構成的に発現するようになるIL-2受容体のサブユニットのうち、我々がクローニングしたサイトカイン受容体コモンγ鎖の発現へのTaxの影響を検討した。γ鎖はほぼすべての血球細胞で発現している。活性化T細胞にIL-2を作用させると一過性にγ鎖の発現は低下し、この現像は転写レベルで起こることを明らかにした。この機構は、免疫反応のため活性化してT細胞を休止状態に戻すのに関与しているのかもしれない。しかし、HTLV-I感染細胞では、IL-2を作用させてもγ鎖の低下はみられない。これは、Taxによりγ鎖の発現が転写レベルで増強され、IL-2による低下分を補っているためであることを証明した。従って、HTLV-I感染細胞での機能的な高親和性IL-2受容体の恒常的な発現は、Taxによるα鎖の発現誘導ばかりでなく、γ鎖遺伝子の転写促進によっても支えられていることになる。γ鎖遺伝子プロモーター領域を単離して、種々のプロモーター欠失変異体で調べたところ、Taxの作用は、転写開始点を+1として、-58から+35の塩基配列の中にあり、この中にはHTLV-I Taxの既知の標的配列は含まれていない。従って、今までに知られていない新たなTaxによる転写促進機構に依っているのかもしれない。
