ウイルス感染病態におけるフリーラジカルによる生体制御機構の解明:特に一酸化窒素ラジカルの関与について

研究概要

本年度は、昨年度に引き続きマウスインフルエンザウイルス肺炎モデルおよびラットヘルペスウイルス(HSV-1)肺炎モデルにおける一酸化窒素(NO)の病態生理学的役割について解析した。その結果、両ウイルス感染モデルにおいて、誘導型一酸化窒素合成酵素(inducible NO synthase, iNOS)の強い誘導が確認され、それぞれの感染局所(マウス肺、およびラット脳)での過剰なNO生成を電子スピン共鳴法により直接照明することができた。一方、インフルエンザウイルス肺炎モデルにおいて、NOとスーパーオキサイド(O_2^-)の相互作用について検討したところ、本モデルにおいてウイルスの肺内増殖によってもたらされるTh1依存性の免疫反応に伴って産生されるinterferon-γおよびtumor necrosis factor-αにより、NOとO_2^-の過剰生成がほぼ同時に誘導されることが明らかになった。同時に、マウス肺内において、NOとO_2^-のすみやかな反応を介してNOやO_2^-よりも科学的反応性に富むパーオキシナイトライト(ONOO^-)の生成がもたらされることを電子スピン共鳴法および免疫組織科学的解析により証明した(Proc.Natl.Acad.Sci.USA 93:2448-2453,1996)。さらに、パーオキシナイトライトの消去剤として最もすぐれているエブセレンを本モデルに投与したところ、致死率の有意な改善が認められた。以上の知見は、ウイルス感染病態においてNOやO_2^-などのフリーラジカル産生により生じるパーオキシナイトライトが、ウイルス感染病態の重要な病原因子として作用していることを示している(Proc.Soc.Exp.Biol.Med.1997,in press)。
