ABO式血液型糖鎖転移酵素遺伝子の法医学的応用と抗原発現の生理学的意義

研究概要

本研究期間では、ABO式血液型抗原系の生理学的意義についてこの点に関して重点的に検討を加えた。これらの抗原が、ヒト、日本猿、ラット及びネコの精巣上体管、前立腺等の雄性生殖器官に発現されており、また、ヒト、日本猿、イヌ、ネコ、ラット、ウサギなどの舌味蕾部位にも発現されていた。これら抗原が種を越えて存在することは、これらの抗原が精子成熟過程や化学的受容器である味蕾細胞において重要な役割を担っていることを示している。また、非分泌型個体においては、ABO式血液型抗原の分泌に明確な民族差が見られることを新たに発見、報告し、非分泌型遺伝子が日本猿等には見られないことから、新しい遺伝子で、民族が明確に分岐された後に生じた可能性を示した。さらに、ヒト胸腺ハッサル氏小体にこれらの抗原の発現を見出し、ABO式血液型抗原の同小体での機能に関して研究継続中である。
法医学的応用面では、ABO式血液型遺伝子型判定用プライマーを用いて、PCR法を行うと、魚類を含めた脊椎動物や家畜、ペット類由来DNAに増幅が認められ、そのパターンにより人獣鑑別の可能性を見出した。このことは、同時に斑痕等を用いたABO式血液型遺伝子型判定時には人獣鑑別が必要になることを示している。また、ABO式血液型のA及びO型糖転移酵素合成遺伝子内のDNA塩基配列多型やその民族差が知られるところとなり、より細かい遺伝子型の簡便な判定方法の開発、民族差を用いた人類学的検討を進行中である。
