胎生期レパトアの機能解析と自己免疫疾患に及ぼすN領域の影響

研究概要	V (variable) 、D (diversity) 、J (joining)の3種の遺伝子の再構成によって形成されたT細胞レセプター(T cell receptor ; TCR)、免疫グロブリン(immunoglobulin ; Ig)遺伝子の可変領域の多様性は、V-D,D-J結合部でのTerminal deoxynucleotidyl transferase (TdT)による塩基付加(N領域)によってさらに増大される。 TdTはagc dependentおよびstage specificな発現調節によりT細胞、B細胞レパトアの形成に多大な影響力を持つが、これがいかなる意味をもつかを明らかにするためジーンターゲティングの手法を用いTdTのプロモーターのみをE_μV_H(IgのV_H遺伝子のプロモーターとH鎖μ領域のエンハンサー)あるいはT細胞特異的なlckプロモーターで置換しTdTの発現パターンを変え、全く新しいT細胞、B細胞レパトアを持ったマウスを作製した。 TdTのプロモーターをE_μV_Hで置換したマウスでは、正常と同程度のTdTの発現を胸腺で見るほか正常では検出できない脾臓、骨髄、リンパ節、胎生期胸腺、胎生期肝臓でも強い発現がみられる。実際IgのIgH鎖、TCRのβ鎖、γ鎖遺伝子の約9割の塩基配列にN領域を認めた。正常ではIgL鎖にN領域はほとんど見られないが、E_μV_Hで置換したマウスのIgL鎖では2/3の塩基配列にN領域を認めた。したがって本来N領域を持たず多様性の制限されたIgL鎖にも新たな多様性を付加できたことになる。これらのヘテロおよびホモ変異マウスの骨髄ではB220^+IgM^+細胞が半分に減少しており、しかも脾臓におけるN領域の頻度は骨髄に比べ明らかに減少していた。この事はIgL鎖にN領域が付加されたB細胞は自己反応生を獲得し骨髄において選択的に除去される可能性を示唆する。胎生期にもTdTを発現するE_μV_H^<+/+>マウスおよびlck^<+/+>マウスは、胎生期レパトアの意義、N領域の胸腺選択に及ぼす影響、種々のγδT細胞の起源及び皮膚、粘膜への生着機序の解明にも役立つものと考える。
