膵臓における熱ショック蛋白質(ストレス蛋白質)の細胞防御機能に関する研究

研究概要

平成9年度に得られた知見は以下の通りである。
1)膵臓におけるHSP発現に及ぼす中枢神経ペプチドの影響
平成8年度までに、膵腺房細胞においてHSP60が細胞防御機構に極めて重要な役割を果たしていることをセルレイン膵炎、L-アルギニン膵炎モデルを用いて明らかにした。全身性ストレスによって膵腺房細胞で誘導されるHSP60の更に詳細な誘導機構を解明するため、ストレス関連神経ペプチドであるCRF,TRHをラットの脳室内に投与し、各種HSPの発現を検討したところ、CRF投与によりHSP60が、TRH投与によりHSP60、72が膵腺房細胞において誘導されることが明らかとなった。これらのことより、膵腺房細胞の内因性細胞防御因子として重要なHSPの発現は、中枢神経系によって制御されている可能性が示唆された。
2)他の消化器系臓器における臓器のHSPの発現、機能に関して
胃粘膜、大腸粘膜、肝細胞における内因性細胞防御因子としての各種HSPに関しても検討したところ、胃粘膜ではHSP72、大腸粘膜ではHSP72,90、肝細胞ではHSP72が細胞防御機構に重要で、膵腺房細胞とは異なることが明らかとなった。
上記1),2)の研究成果より、in vivoでは臓器間で細胞防御機構に重要なHSPの種類が異なることが明らかとなり、膵腺房細胞においては、HSP60を特異的に誘導することが、治療への応用を考慮した場合には効果的であることが示唆された。よって、1)で明らかにした、HSP60の特異的な誘導法は特に重要であると考えられた。
