肝内マクロファージ(Kupffer細胞)由来活性酸素による肝癌細胞障害機序の解明

研究概要

昨年度に引き続き本年度においては、Kupffer細胞によるNO・TNF-α放出動態と肝癌細胞障害・アポトーシスとの時間的空間的相関およびその制御機構を解明してきた。すなわちKupffer細胞と肝癌細胞の共培養系におけるNOおよびTNF-α産生動態の定量と、肝癌細胞内ミトコンドリア呼吸能の変化、細胞膜・核膜障害、DNA fragmentation化を共焦点レーザー顕微鏡下に可視化、定量するシステムを確立した。NO・TNF-α合成系の関与を裏付けるためにinducible NO synthase(iNOS)mRNAおよびTNF-α mRNAに対するアンチセンスプローブを用いて抑制実験を行なったほか、fluorescence in situ hybridization(FISH)法を応用し培養細胞内iNOSおよびTNF-α mRNAの誘導を確認した。さらに各種接着因子(ICAM-1、CD18等)に対するモノクローナル抗体を用いて上述の実験系に対する抑制実験を行った。以上の結果、肝癌細胞とのICAM-1、CD18を介した接着によりkupffer細胞のNO・TNF-α合成が増加し、肝癌細胞にnecrosisのみならずapoptosisが惹起されることが明らかとなった。さらに共焦点レーザー顕微鏡的観察と組み合わせたFluorescence in situ DNA-protein結合法の開発により活性化した転写因子(NF-κB)がKupffer細胞におけるiNOS mRNAおよびTNF-α mRNAの誘導に関与していることを証明することに成功した。
