硫酸基糖転移酵素の発現クローニングと癌転移との関連についての研究

研究概要	血液凝固系関連物質や細胞間接着物質との相互作用が注目される硫酸化糖鎖の合成を司る糖転移酵素のクローニングを目指し、平成8年度の研究計画に従い、以下の研究を行った。 1.cDNAライブラリーを用いた硫酸化糖鎖の発現 SV40 T抗原産生細胞COS細胞表面には、目的とする硫酸化糖鎖の前駆体である構造の存在をモノクローナル抗体を用いた抗原解析及び糖脂質抽出にて認めた。真核細胞発現ベクターを用いて作製した、硫酸化糖鎖抗原陽性のHepG2細胞、及び肺小細胞癌株のcDNAライブラリーを、COS細胞にトランスフェクションを行った。硫酸化糖鎖特異的モノクローナル抗体を付着させたプレート上に、トランスフェクタント細胞を反応させ、Hirt法によるplasmid DNAの回収を行った。この一次Panningを2ク-ル施行した。以後、Panning法を繰り返し、特異クローンを求めていき、その中で硫酸基糖転移酵素活性を発現するクローンの同定を行う。 2.糖脂質特異的糖転移酵素との類似性末端ガラクトースに硫酸基の結合した構造の中で、最も単純な構造(SM3)の合成に関与する、糖脂質特異的硫酸基糖転移酵素が先日報告されたが、2型膜糖蛋白構造のそのカルボキシル末端側のアミノ酸部に硫酸基基質と反応する部位が存在すると考えられ、その部分のdegenerative核酸配列を合成した。上記のcDNAとそのdegenerative primersを用い、RT-PCRを施行し、数種の産物を得た。以後、得られたPCR産物の核酸配列を決定し、糖転移酵素をコーディングしている可能性のあるものに関して、得られているcDNAライブラリーとのColony-Hybridizationを行う。を行なう。
