パラインフルエンザウイルス4型感染症の分子疫学的研究

研究概要

1。三重大学・小児科などから集めた患者血清約250検体をELISA法にて解析したところ約20検体が陽性または偽陽性であった。またウイルス中和反応においても同じ血清がウイルスの増殖を抑制した。ただし4A、4Bのどちらを抑制しているかの区別は不可能であった。共通抗原を持つと思われる2型ウイルスなどの交差反応の検討はこれからである。
2。4型ウイルス特異的抗原作製のため、4型ウイルスのHN蛋白を発現しているL929細胞を樹立した。
3。4Aウイルスと4Bウイルスの抗原性の違いは、HN蛋白によることが、ELSISA法により明らかとなっている。そこで感染が4Aウイルスあるいは4Bウイルスによるのか区別するため、また抗原性の違いの原因部位を探るため、4BのHNの糖鎖結合部位2ヵ所にpoint mutationを導入し、4AのHNにちかいクローンを作製し発現細胞を樹立した。
今後これらの細胞を使用し、感染の実態および抗原性の差の解明を行う予定である。
