アルツハイマー病におけるCa^<2+>非依存性novelプロテインキナーゼC異常

研究概要

タンパクのリン酸化は細胞の機能および生存維持に極めて重要な現象である。申請者らは、すでにリン脂質依存性Ca^<2+>依存性セリン/スレオニンタンパクリン酸化酵素であるconventionalプロテインキナーゼC(cPKC)のアルツハイマー病脳における変動について解析した。PKCにはcPKCの他、キナーゼ活性化因子としてCa^<2+>を必要としない新しいタイプのnovel PKC(nPKC)が存在していることが判明してきた。本研究では、アルツハイマー病におけるnPKC異常について検討した。
Ca^<2+>非依存性PKC活性は、大脳皮質(側頭葉)組織の膜分画、細胞質分画のいずれにおいてもアルツハイマー病群で対照群の約50%に低下していた。Ca^<2+>依存性PKC活性は、細胞質分画において有意に低下していた。nPKCの3種類のアイソザイム(δ,ε,ζ)に対する特異抗体を用いたWestern blot解析を行った。PKC-δは75kDaタンパクとして、PKC-εは90kDaタンパクとして、PKC-ζは70kDaタンパクといてヒト脳組織に発現していた。アルツハイマー病脳の膜分画および細胞質分画のいずれにおいてもPKC-εのタンパクレベルが対照群に比べ有意に減少していた。PKC-δおよびPKC-ζには変動は見られなかった。PKC-εの特異抗体による免疫染色を行った。PKC-ε免疫活性は、海馬に特に強く、主にニューロン発現し、グリアには発現していなかった。細胞内では、細胞質内に局在し、核には存在していなかった。
本研究で得られたアルツハイマー病脳におけるnPKCおよびcPKC活性の低下は、amyloid precursor protein(APP)の代謝過程に変動を及ぼし、分泌型APPの減少、可溶性amyloid betaタンパクの増大をもたらし老人斑の形成機序に関与している可能性がある。また、PKC-εの減少はニューロンの変性機序のみならず、痴呆発現などアルツハイマー病でみられる高次脳機能障害の一因である可能性が示唆された。
