神経栄養因子BDNFのヒト脳での局在と神経変性疾患での病態に関する組織化学的研究

研究概要	抗BDNF抗体を作製し特異性を検定した。本抗体はドットブロットで3.91pmolの抗原を認識できた。ラット脳ホモジェネートを用いたイムノブロットではBDNFモノマーの分子量に相当する約14kd付近に単一バンドを認めた。以上から、本抗体はラット脳内のBDNFを特異的にかつ高感度に認識すると考えた。本抗体を用いて、ラット脳でのBDNFの免疫組織化学的局在を検討した。その結果、BDNFはラット脳では従来考えられていたよりも広範に分布していること証明した。BDNFは神経細胞、樹状突起、神経終末の局在し、陽性神経細胞は終脳から脊髄に分布していた。大脳皮質と海馬では錐体細胞、介在神経細胞が染色された。神経細胞体内では顆粒状に染色された。一部の神経細胞の表面に神経終末と考えられるドット状の反応産物を認めた。また、主として第1層に陽性線維を多数観察した。その他、以下の諸核に陽性神経細胞を認めた。前脳基底部、線状体、扁桃体、視床下部、視床内側核群、黒質、脳神経運動諸核、橋核、オリーブ核、脊髄前角細胞などである。ただし、陽性反応性は諸核によりその程度は異なり、同一神経核内でも細胞によりことなっていた。例えば、視床下部では室傍核と視索上核に特に強い陽性反応を認めたが、大型神経細胞のみが染色され、その他の小型、中型神経細胞はほとんど染色されなかった。以上から、BDNFがラット脳内の広範な領域で特定の神経細胞群において栄養因子として作用していることが示唆された。また、BDNFが神経終末と神経細胞表面に存在することから、BDNFが軸索輸送され、神経終末を介して取り込みが行われることが推測された。さらに、本抗体をもちいて筋萎縮性側索硬化症剖検例の脊髄を免疫組織化学的に検索し、その前角を中心にBDNF陽性線維の増加を観察した。この所見はBDNFが本疾患においてその代謝に変化を生じていること示唆する。
