HTLV-I感染によるミクログリア・アストログリアの反応と神経細胞死のメカニズム

研究概要	初代培養による小脳顆粒細胞に対するHTLV-I感染細胞株であるMT-2、HCT-I、W7TM-I、HAM患者より分離したT-cellなどの培養上清の細胞障害性を、MTT法、fluorescin diacetate法による生細胞数の測定を行ったがMT-I、MT-2で神経細胞障害性が容量依存性に見られた。この現象はtax遺伝子の関与が推測されたがtax蛋白に対する抗体を含んだ患者血清での中和実験では、抑制できずtax蛋白そのものというよりはtax蛋白によって発現が亢進したサイトカインなどの因子によるものと推測された。次ぎに、初代培養のアストロサイト、ミクログリア、小脳顆粒細胞など神経系細胞へのtax蛋白遺伝子導入を試みたが、導入効率で問題があった。そこでミクログリアと基本的に同じ性格を持つと考えられる細胞としてU937細胞株を用い遺伝子導入を試み、良好な導入効率がえられた。tax遺伝子をサイトメガロウイルスのプロモーターの顆粒に挿入したvectorを用いてリポフェクトアミン法でU937細胞株に導入し、神経系に障害性を持つnitric oxide(NO)を測定、またinducible NO synthase(iNOS)のmRNA発現量を半定量PCR法で測定した。その結果、tax遺伝子を導入したU937細胞はNO濃度は高値で、iNOSのmRNAは有意に多く発現しておりIFN-γ刺激によって更に増強された。このことから、HAMの発症病理に於てresidentialなマクロファージやミクログリアより誘導されるNOが神経組織障害に関与している可能性が示唆された。これまでにHAMの唯一のモデルであるいわゆるHAMラットではマクロファージ/ミクログリアの組織内での障害への関与が示唆されており我々の研究結果と合わせてHAMの発症病態に重要と思われる。
