エンドセリン変換酵素の調節機構ならびに病態モデルでのその発現の変化

研究概要

エンドセリン変換酵素(以下ECE-1)は、エンドセリンの産生過程において、Prepro-ET-1より産生された中間体Big-ET-1をET-1に変換される役割を有する酵素である。私達はエンドセリン変換酵素の発現調節に関し検討し、以下の実験結果を得た。
(1)ECE-1mRNA発現調節
ウシ大動脈培養内皮細胞、ならびにラット大動脈培養平滑筋細胞において、それぞれの細胞におけるECE-1mRNA発現を、ウシECE-1 cDNAより作成したプローブを用い、Northern blotting法により検討した。実験においては、各培養細胞がcpnfluentな状態で、無刺激無血清培養下24時間における発現量をコントロールとして、種々の因子による刺激後の発現量の変化を検討した。
1。内皮細胞:TGF-β 3ng/ml、TPA 100μM、ATPγS10μM、Dibutyl cAMP 10μM刺激後1、4、12時間におけるECE-1 mRNAの変動はコントロールに比べて最大2倍以下にとどまった。
20平滑筋細胞:TGF-β 3ng/m、Angiotensin 1μM、TPA 100μM刺激に対し有意なECE-1 mRNAの変化は認められなかった。
(2)各種循環器疾患の病態モデルにおける心血管系でのECD蛋白発現の変化
家兎免疫抗ウシECE polyclonal抗体を用い免疫組織化学を行なった。
1。ラット頚動脈balloon傷害内膜剥離、血管壁肥厚モデル:内膜肥厚部に一致し、内皮細胞のみならず増殖平滑筋細胞においてもECE-1蛋白発現を認めた。
2。モノクロタリンによるラット肺高血圧、右心不全モデル:Western blotting法による検討において、心室におけるET-1蛋白の発現は右心不全ラットではコントロールラットに比べて増加していたが、ECE-1蛋白の発現もやや増大の傾向を認めた。
以上よりECE-1は各細胞においてconstitutiveに発現しており、その遺伝子発現は比較的安定であるが、生体内においては病態により発現量が変化する可能性があると考えられた。
