原発性免疫不全症候群の病因遺伝子の解明に関する研究

研究概要

原発性免疫不全症候群には多彩な病型が見られ,その病因・病態も複雑多岐にわたる。本研究の目的はこれらの種々の病型の病因遺伝子を明らかにし、個々の患者の遺伝子の異常や病態を明確にすることにより原発性免疫不全症候群を系統的に整理分類し、治療、予防に寄与することである。さらにヒト免疫機構の解明にも寄与する。
その研究の結果、本年度は主として以下の点が明らかになった。
Common variable immunodeficiencyのうちIgGとIgAの低下を示す型では免疫グロブリン遺伝子重鎖Iγ領域の核クロマチン構造がopenになりにくいことによりこの病型が発症していることが明らかになった。さらに全クラスの免疫グロブリンの低下を示すCommon variable immunodeficiencyではsurrogate light chainの発現低下やプロティンキナーゼCの反応が低下していることも明らかになった。
IgG2欠損症ではインターフェロンγの産生低下がその病因に大きく関わっていることが明らかになった。
Bloom症候群については病因遺伝子(BLM遺伝子)が明らかにされ、cDNAの全長4437bpでアミノ酸1417をコードしていることが明らかにされた。1兄妹例ではCAAの欠失がありstop codonによりアミノ酸185の長さの短いタンパクが作られていることが示唆された。また、発癌との関連でp53遺伝子の役割も明らかになった。
