インスリン分泌制御におけるPI3キナーゼの役割の解析

研究概要	我々が樹立した膵β細胞体MIN6はグルコースに反応性のインスリン分泌を示す。細胞内シグナル伝達に重要な役割をもつと考えられるphosphatidylinositol 3-kinase(PI-3K)とミオシン軽鎖リン酸化酵素(MLCK)の阻害剤であるwortmanninを用いてMIN6細胞からのインスリン分泌を検討し、その結果に基づきインスリン分泌制御におけるMLCKとPI-3Kの役割を詳細に検討した。まず、MIN6細胞を培養し、2日後にさまざまな濃度(1nM〜10μM)に希釈したwortmannin中で処理し、15mMグルコースの存在下でのインスリン分泌を測定した。インスリン分泌と最も増加させる濃度のwortmanninでMIN6細胞を処理した後、2〜25mMグルコースに反応性のインスリン分泌を測定した。その結果、高い濃度ではインスリン分泌が阻害されるが、低い濃度では逆にグルコース依存性インスリン分泌が顕著に増加することが示された。グルコースによる細胞内Ca^<++>の増加はwortmanninに影響されなかった。wortmannin処理によりPI-3K活性は顕著に低下していた。wortmanninで処理したMIN6細胞を、抗wortmannin抗体で免疫沈降し、western blotにより解析したところ、供沈される蛋白がPI-3KあるいはMLCKと一致した。MIN6細胞をIGF-1で刺激するとインスリン分泌が大きく低下したが、これはwortmanninにより回復した。これらの結果から、MLCKはインスリン分泌に直接関与していること、PI-3Kはβ細胞では分泌したインスリンからのnegative feedbackのシグナルを伝えるのに関与していることが推定された。
