研究概要

1:甲状腺ホルモンレセプター結合因子であるNuclear receptor co-repressor(N-CoR)のvariantのクローニング
既に酵母発現系を用いるTwo hybrid systemにより1つの甲状腺ホルモンレセプター結合因子(B1)を得、塩基配列決定によりmouse nuclear receptor co-repressor (mN-CoR)のhuman homologue (hN-CoR)であった。B1の全長をクローニングするためHela細胞より構築されたcDNAライブラリーをPlaque hybridization法にてスクリーニングしたところ、N-CoRと約80%の相同性を認めるN-CoRvariantであるクローンH3を得た。このクローンH3より演繹されるアミノ酸配列は、N-CoRのアミノ端が欠失し、またそのカルボキシル端の120個のアミノ酸が異なる55個のアミノ酸に置換したhN-CoRの新たなvariantと考えられるものであった。mN-CoRは、TR、レチノイン酸レセプター(RAR)と結合し、その転写活性抑制に関与するco-repressorとされ、mN-CoRのアミノ端に転写活性抑制に関する部位が、またそのカルボキシル端にTR,RARと結合する部位があるとされている。variant H3では、N-CoRがレセプターと結合するのに必要なカルボキシル端の部位の一部のアミノ酸が置換されていることより、TR,RARとの結合が障害されているか、レセプターとの結合特異性が変化している可能性が考えられた。
2:N-CoR isoformの組織発現の検討
hN-CoRとそのvariantであるH3の細胞における発現を、様々なヒトの細胞株のtotal RNAを用いてRT-PCR(reverse transcription-polymerase chain reaction)を行うことにより検討した。検討した10種類の細胞株すべてにおいてhN-CoRとvariant H3の発現を認めた。
