アポトーシスによる膵β細胞死とBcl-2ファミリーによるその制御

研究概要	本研究は、(1)アポトーシスによる膵β細胞死の解明、および、(2)最近新しく発見されたアポトーシス抑制蛋白Bcl-2による膵β細胞死の制御、の2点を目的として行った。まず、膵内分泌細胞株(βTC1、αTC1、NIT-1)において、サイトカイン(IFNγ、IL-1β、TNFα)により、アポトーシスに特徴的なDNAの断片化が経済的に増加した。この成績は、膵内分泌細胞において、アポトーシスを介した細胞死が誘導され得ることを示す。断片化DNAの量は、βTC1>NIT-1>αTC1の順であった。これらの細胞株において、アポトーシス抑制蛋白であるBcl-2の発現をみると、βTC1<<NIT-1≒αTC1であった。次に、上記膵内分泌細胞株のうち、Bcl-2蛋白の発現が最も少量で、しかもアポトーシスが最も誘導されやすかったβTC1細胞にbcl-2遺伝子をトランスフェクトし、過剰発現させた。その結果、Bcl-2蛋白の過剰発現したβTC1細胞では、対照のβTC1細胞に比較して、断片化DNAの量が約半分に減少した。以上の成績より、膵β細胞がサイトカインによってアポトーシスの機序を介して死に得ること、また、そのβ細胞死をBcl-2によって抑制し得ることが明らかになった。
