Estramustineのmalignant gliomaの成長に対する抑制効果

研究概要

(1)[^3H]-thymidine up takeとMTT assayによりglioma cell bireで時間及び濃度依存性に細胞増殖を抑制した.しかし正常astrocy Teでの抑制効果はみられなかった。
(2)in vitro Matrigel assayでは細胞侵潤は抑制された。従って正常な脳組織での腫瘍細胞侵潤中抑制されうると示唆している。
(3)collagemolysis assayではCollagenase活性の低下がみられた。これは(2)の実験においてEstramustineはcollagenase(matrix proteinase)活性の低下をおこす事を示していた。
(4)Zymogramとin situ hybridizationによりmatrix metallo proteinase分泌が抑制されていた。従って基質の溶解が(2)において抑制されたものと思われた。
以上よりEstramustineは細胞の増殖抑制作用があるばかりではなく腫瘍細胞の遊走と組織侵潤についても抑制効果がありその一つがMatrix metallo proteinaseの分泌抑制である。これらの結果を昨年Nourosurgeryでの論文発表とヨーロッパ脳腫瘍学会で発表した。
