尿路感染症成立機序の解明(分子生物学的アプローチによる)

研究概要

細菌の線毛は大別するとマンノース感受性(MS)線毛とマンノース抵抗性(MR)線毛に分けられる。我々は、セラチアよりMR線毛及びMS線毛の遺伝子のクローニングを行い、ラットの腎孟腎炎モデルに於いてMS線毛保有菌がMR線毛保有薗より腎瘢痕形成能が強いことを示した。in vitroで、MS線毛保有菌は、MR線毛保有菌に比し、ヒト白血球に貪食され易くSuperoxideの産生刺激が強いことが判明した。以上よりMS線毛が感染部位で白血球のSuperoxideの産生を促し組織破壊が惹起されるものと思われる。MS線毛は、いくつかのsubunitより構成されているが、そのうち線毛の先端に存在するAdhesin蛋白が血球凝集に関わっていることが知られている。そこで大腸菌のMS線毛のAdhesin遺伝子の欠失mutantを作製した。このmutantは線毛を有するが血球凝集能を示さなかった。このAdhesinを欠くmutant線毛を有する菌とwild typeのMS線毛を有する菌をラットの腎に接種するとwild typeのMS線毛を有する菌は、腎瘢痕形成を強く惹起するが、mutantは殆ど腎瘢痕形成を惹起しなかった。このことより、MS線毛のAdhesinが腎孟腎炎におけるはん痕形成に重要な役割を果たしていることが示された。mutant線毛を有する菌はwild typeのMS線毛を有する菌と同程度ヒト白血球に貪食されやすいが、superoxideの産生刺激は低かった。以上より線毛のAdhesinが白血球のsuperoxide産生を強く刺激し、線毛を介する貪食は他のsubunitも関与していると考えられ、このことは新しい知見である。これまでの研究で腎瘢痕を引き起こす宿主側および細菌側の因子の解明がかなりの程度なされ、また細菌側の重要な因子であるAdhesinの同定ができた。
