プロスタグランジンE受容体の機能とその生理的役割に関する分子生物学的研究

研究概要

1、EP3アイソフォームによるG蛋白質活性化調節機構の解析 マウスEP3には、C末端鎖の構造のみの異なる3つのアイソフォーム(EP3α、β、γ)が存在する。このC末端鎖のG蛋白質活性化における役割を明らかにする目的でC末端鎖を欠損した変異受容体を作製した。変異受容体は、常時アゴニストに依存しないGi活性を示し、アゴニスト依存性の活性化は全く見られなかった。そこで、3つの各アイソフォームのアゴニスト非依存性のGi活性を調べると、EP3βは全く非依存性の活性は示さなかったが、EP3αは約半分が非依存性の活性化型であり、EP3γはその殆どが非依存性の活性化型で存在した。以上のことから、EP3のアイソフォームはアゴニスト非依存性の活性において異なっており、C末端鎖はEP3本体が示すGi活性をアゴニスト依存的に抑制する機能がある。その強さはC末端鎖の構造によって異なり、βのC末端鎖はもっとも強い抑制を示し、次いでαのC末端鎖、そしてγのC末端鎖には殆ど抑制活性が見られない。
2、PGE受容体サブタイプの中枢神経系における発現分布 機能的に異なる4つのPGE受容体サブタイプ、EP1、EP2、EP3、EP4の中枢における発現分布をin situ hybridization法により解析した。4つのサブタイプの中で、EP3は最も発現量が高くかつ幅広く分布していた。その発現はニューロンに特異的で、特に海馬や視床下部、脳幹では強い発現を持つニューロンが点在し、大脳皮質や視床では中程度の発現を持つニューロンが多く見られた。これに対して、EP4は視床下部と脳幹の一部の核に、EP1は視床の一部の核に発現が認められた。一方、EP2はどこにも発現は見出されなかった。この様に、各サブタイプは中枢において異なる部位に発現し、神経活動を制御していることが示唆された。
