研究概要	大腸菌のスペルミジン優先取り込み系は基質結合蛋白質(PotD)、ATP結合部位を有する膜表在性蛋白質(PotA)、及び2種のチャンネルを形成する膜貫通蛋白質(PotB及びPotC)より構成されるABCtransporterの一種である。このうちのPotAとPotD蛋白質を均一に精製し、酵素化学的性質を検討すると同時に、site-directed mutagenesisによりアミノ酸置換蛋白質を作製しその性質を検討した。1)精製したPotA蛋白質はATPase活性を示し、その比活性は400nmol/min/mg proteinであり、基質ATPのKm値は385μMであった。ポリアミン輸送欠損株NH1596のPotA蛋白質は135番目のバリンがメチオニンに変異しており、ATPase活性を欠損していた。この系によるポリアミン取り込みがSH阻害剤により阻害されることから、PotA蛋白質中に存在する3個のシステイン残基(C26、C54及びC276)を変異させた結果、54番目のシステイン残基が活性発現に必須であった。従って、PotA蛋白質はN末端側にATPaseの活性中心が存在することが明らかとなった。2)PotD蛋白質のX線結晶解析より、ポリアミン結合に関与するアミノ酸残基が同定されたので、それらを変異させたPotD蛋白質を作製し、ポリアミン結合活性と輸送活性を測定した。その結果、結合活性と輸送活性はほぼ相関しており、4分子の酸性アミノ酸残基(E36、D168、E171、D257)、3分子のトリプトファン残基(W34、W229、W255)、及び3分子のチロシン残基(Y37、Y85、Y293)が基質スペルミジンとの相互作用に関与していた。特にスペルミジンのジアミノプロパン部分と相互作用するE171、W255及びD257の変異により活性が著しく低下することが明らかとなった。
